I think it would be a nice feature to have the option to pass custom variables into the Send replies to form field. I only say this because it was a request from a client and I'm pretty sure that is not a feature of CM. Opinions?
Hey there jeremypeter, thanks for the suggestion! While we don't specifically have a "reply-to" address custom field (with a pre-filled value, anyway), you can certainly use custom fields to add an email address to your email content. Note that it's likely that many readers will simply hit "reply" on a newsletter, so it's important that you have a valid reply-to email address to start off with.

Yes, excellent idea. This is quite different than using custom fields in the email content. Allowing the ability to dynamically pull the from/replyto info from ist data would add a level of personalization that can only be done currently via the API. A use case, would be a send from a list containing data with associated sales reps, where you would like to have the email coming from the sales rep.
